Description

2-YT Broth is a nutrient-rich microbial growth medium used for cultivation of E. coli and growth of hosts for replication of M13 vectors. The 2-YT formulation is similar to Lennox LB medium, but contains double the concentration of yeast extract and a 60% higher concentration of peptone. 2-YT medium allows cells to reach higher density and increase recombinant protein production compared to LB media formulations.

2-YT Broth formulation
Formulation per liter: 16 g SELECT Peptone 140 (pancreatic digest of casein), 10 g SELECT Yeast Extract, and 5 g NaCl.
Note: To prepare 1 L of 2-YT medium, weigh 31 g of powder and add distilled water to 1 L. Adjust the pH value to 7.0 with 5 N NaOH and sterilize by autoclaving for 20 minutes at 121°C.
